Our research focuses on 3D reconstruction of biological macromolecules by single particle cryo electron microscopy. In this technique, a large number of unstained molecules are imaged as projections of unknown orientation. By computational means, their relative orientations can be determined and the underlying 3D structure retrieved. Much of our efforts concentrate on the development of improved methods for image processing in order to routinely obtain higher resolution and improved analysis of structural heterogeneity. To this end, we have developed our own platform for image analysis (Strul/RAD/Volrec). The present focus is on implementation on and evaluation of software for automatized image acquisition and high-throughput reconstruction.
We are involved in a number of collaborations which incorporate structural analysis by these techniques:
-Structure and conformational changes in bchid magnesium chelatase (J. Lundqvist, S. Al-Karadaghi, M. Hansson (Biochemistry LU))
-Iron storage and transport by frataxin (S. Al-Karadaghi)
